چکیده

Polymorphism. TaqI restriction fragment length polymorphism (RFLP) in the stearoyl-coenzyme A terminal desaturase ( SCDl) gene of purebred Japanese Black cattle. Source and Description of Probe. A 2.7-kb cDNA insert was excised from a pSp86 clone by digestion with EcoRI and ScaI. The insert contains the complete coding sequence of the rat SCDl gene (Thiede et al., 1986). Method of Detection. Prehybridization solution contained 5x SSC buffer, lox Denhart's solution, .05 M Nap04 buffer (pH 6.7), .5% SDS, 5% dextran sulfate, 50% formamide, and 300 pg/mL salmon sperm DNA. The hybridization solution contained l x Denhart's, .02 M Nap04 buffer (pH 6.71, .5% SDS, 10% dextran sulfate, 50% formamide, and 100 pg/mL salmon sperm DNA.
